To investigate the mRNA expression regulated by Setdb1, we established AML12 knockdown cell model by infecting Setdb1 siRNA for 72 hours Overall design: We then performed gene expression profiling analysis using data obtained from RNA-seq of 2 different cell groups (siNC vs siSetdb1, 3 samples per group).

为探究受Setdb1（SET结构域分叉蛋白1）调控的mRNA表达水平，我们通过感染Setdb1小干扰RNA（small interfering RNA, siRNA）并培养72小时，构建了AML12基因敲低细胞模型。实验整体设计：随后我们基于两组不同细胞（阴性对照siRNA组siNC与Setdb1靶向siRNA组siSetdb1，每组各3个生物学重复样本）的RNA测序（RNA-seq）数据开展基因表达谱分析。
