16s rRNA sequencing to assess the effect of 2H-GPS on gut microbiota composition of Alzheimer's mouse model

In our previous study, we found that gentiopicroside isolated from Gentiana rigescens Franch has a significant antiaging activity and regulates mitophagy and oxidative stress. On the basis of these studies, several compounds were synthesized and evaluated for neuroprotective activity with PC12 cells bioassay and yeast lifespan assay, and 2Hgentiopicroside was selected for further study. The result in this study show that 2HGPS improves the symptoms of AD mice through combination of the regulation of Wnt signaling pathway and the microbiota gut brain axis, and the mechanism of action of 2H-GPS is distinct from that of Done.
在我们此前的研究中,我们发现从坚龙胆(Gentiana rigescens Franch)中分离得到的龙胆苦苷(gentiopicroside)具有显著的抗衰老活性,且可调控线粒体自噬与氧化应激。基于前述研究成果,我们合成了多种化合物,并通过PC12细胞生物测定与酵母寿命试验评估其神经保护活性,最终筛选出2H-龙胆苦苷(2Hgentiopicroside)开展后续研究。本研究结果显示,2HGPS可通过协同调控Wnt信号通路(Wnt signaling pathway)与肠道菌群-肠-脑轴(microbiota-gut-brain axis)改善阿尔茨海默病(AD)小鼠的病症,且2H-GPS的作用机制与Done存在显著差异。
