Genetic variations play an important role in tumor development and metastasis. Hepatocellular carcinoma (HCC) is one of leading cause of cancer-related death. Despite improvements in surveillance and clinical treatment strategies, the prognosis of HCC remains dismal. Affymetrix SNP 6.0 array were used to evaluate the genetic characteristics of tumor DNA in 30 HBV-related HCC patients who were underwent liver transplantation. Recurrence related SNPs were selected and validated. Overall design: Affymetrix SNP 6.0 arrays were performed according to the manufacturer's directions on DNA extracted from formalin-fixed paraffin-embedded HCC tissues
遗传变异在肿瘤发生与转移过程中发挥重要作用。肝细胞癌(Hepatocellular carcinoma, HCC)是癌症相关死亡的主要诱因之一。尽管当前监测手段与临床治疗策略已有所改善,但肝细胞癌的预后仍不容乐观。本研究采用Affymetrix SNP 6.0基因芯片(Affymetrix SNP 6.0 array),对30例接受肝移植的乙型肝炎病毒(Hepatitis B Virus, HBV)相关肝细胞癌患者的肿瘤DNA遗传特征进行评估,并筛选验证了与复发相关的单核苷酸多态性位点(Single Nucleotide Polymorphism, SNPs)。
实验设计概况:本研究严格按照制造商提供的操作指南,对从福尔马林固定石蜡包埋的肝细胞癌组织中提取的DNA开展Affymetrix SNP 6.0基因芯片检测。
