Gene expression profiles of MLL-AF9 leukemia cells in C57BL/6 mice and Rag2-/- mice

The relative importance of T/B lymphocytes and NK cells during acute myeloid leukemia (AML) progression has been unclear. In this study, we compared gene expression profiles of mouse AML cells expressing MLL-AF9 collected from immunologically normal wild-type (WT) C57BL/6 mice, Rag2-/- mice lacking mature T and B cells, and NSG mice lacking T, B and NK cells. MLL-AF9 leukemia cells coexpressing GFP were transplanted into immunologically normal C57BL/6 mice, Rag2-/- mice lacking mature T and B cells, and NSG mice lacking T, B and NK cells. GFP+ leukemia cells were collected from bone marrow and isolated using cell sorter before RNA extraction.
急性髓系白血病(acute myeloid leukemia, AML)进展过程中,T/B淋巴细胞与自然杀伤细胞(NK cells)的相对重要性此前尚未明确。本研究对比了取自三类小鼠的表达MLL-AF9的小鼠AML细胞的基因表达谱,这三类小鼠分别为免疫正常的野生型(wild-type, WT)C57BL/6小鼠、缺乏成熟T、B细胞的Rag2-/-小鼠,以及缺乏T、B与NK细胞的NSG小鼠。将共表达绿色荧光蛋白(green fluorescent protein, GFP)的MLL-AF9白血病细胞,移植至上述三类免疫背景的小鼠体内。从骨髓中采集GFP阳性(GFP+)白血病细胞,通过细胞分选仪分离后进行RNA提取。
