The goals of this study is to compare the differently expressed genes in abdominal aorta tissue of WKY and SHR as well as differently expressed genes in the abdominal aorta tissue of SHR with or without neferine treatment. The rat (n=15) were randomly divided into 3 groups: WKY,SHR, and SHR + neferine - H (high concentration) groups (n=6 for each group). Rat in WKY and SHR groups were intragastrically with double distilled water (dd H2O); while rat in SHR + SHR + neferine - H groups were intragastrically with 10mg/kg/D of neferine for 10 weeks. Then the abdominal aorta were used to identify differentially expressed genes among different groups. RNA sequencing was performed with abdominal aorta of mice in 3 groups: WHY, SHR, SHR + neferine - H treated (n=5 for each group) to identify the differently expressed genes and related pathway.
本研究旨在比较Wistar-Kyoto(WKY)大鼠与自发性高血压大鼠(SHR)腹主动脉组织中的差异表达基因,以及经莲心碱(neferine)处理与未处理的SHR大鼠腹主动脉组织中的差异表达基因。将15只大鼠随机分为3组:WKY组、SHR组及SHR+高浓度莲心碱(neferine-H)组,每组6只。WKY组与SHR组大鼠经灌胃给予双蒸水(dd H₂O);SHR+neferine-H组大鼠则以10mg/kg/日的剂量灌胃给予莲心碱,连续干预10周。随后采集各组大鼠的腹主动脉组织,用于鉴定不同组间的差异表达基因。此外,对3组大鼠的腹主动脉组织进行RNA测序,每组5只,以鉴定差异表达基因及其相关通路。
