Study investigated the efficiency of 1,4-dioxane degradation by DD4 in two groundwater sample in landfill leachate site located in Pennsylvania, USA. The site contaminated 1,4-dioxane concentration is as low as tens of ppb, which is completely degraded by inoculated strain DD4 using propane and 1-propanol as auxiliary substrate. For community analysis v3-v4 region of bacteria was amplified using 338F and 806R primers. The resulting amplicons were sequenced using Illumina MiSeq sequencing technology. Microbial community diversity was analyzed before and after augmentation with DD4 in both samples MW-A and MW-B with different dioxane(onsite) concentration.

本研究以美国宾夕法尼亚州某垃圾渗滤液污染场地的两份地下水样品为研究对象,考察了DD4对1,4-二氧六环(1,4-dioxane)的降解效能。该场地的1,4-二氧六环污染浓度低至数十ppb,接种以丙烷与1-丙醇为辅助底物的菌株DD4可将其完全降解。针对微生物群落分析,采用338F与806R引物对细菌的v3-v4可变区进行扩增,所得扩增子通过Illumina MiSeq测序技术完成测序。针对两份现场1,4-二氧六环污染浓度各异的样品MW-A与MW-B,分别在接种DD4前后对其微生物群落多样性进行了分析。
