Hepa1-6-bearing C57BL/6 mouse model was established to evaluate the therapeutic efficacy of Ganoderma lucidum extract (GLE) in HCC. The GLE treated and untreated mice were used for transcriptioanl profiles detection, and 126 differentially expressed lncRNAs and 558 DE mRNAs were identified, respectively. The bioinformatics analysis shows that the GLE could suppress the growth and proliferation of HCC by PI3K/Akt/mTOR and MAPK signaling pathway, and also could induce apoptosis of tumor cell by mitochondrial and death receptor pathway. 本研究构建了携带Hepa1-6细胞的C57BL/6小鼠荷瘤模型,用于评估灵芝提取物(Ganoderma lucidum extract,GLE)对肝细胞癌(Hepatocellular carcinoma,HCC)的治疗效果。将经GLE处理与未处理的小鼠进行转录组谱检测,分别鉴定出126个差异表达长链非编码RNA(DE lncRNAs)与558个差异表达信使RNA(DE mRNAs)。生物信息学分析结果表明,GLE可通过PI3K/Akt/mTOR信号通路与丝裂原活化蛋白激酶(Mitogen-activated protein kinase,MAPK)信号通路抑制肝细胞癌的生长与增殖,同时可通过线粒体途径与死亡受体途径诱导肿瘤细胞凋亡。实验设计:共纳入6份样本,包括3只经GLE处理的Hepa1-6细胞荷瘤C57BL/6小鼠与3只未接受GLE处理的Hepa1-6细胞荷瘤C57BL/6小鼠,用于相关分析。
