EpiSC-5 mouse epiblast stem cells were transduced with lentiviral vectors carrying shRNA for MR candidate genes or non-targeting controls. Expression profile was obtained 72h after transduccion. EpiSC-5 mouse epiblast stem cells were transduced with lentiviral vectors carrying shRNA for candidate pluripotency regulators on on-targeting controls. mRNA was isolated 72h after transduccion and profiled using PLATE-Seq (PMID 28740083).

将EpiSC-5小鼠上胚层干细胞(EpiSC-5 mouse epiblast stem cells)采用携带针对MR候选基因的短发夹RNA(shRNA)或非靶向对照的慢病毒载体(lentiviral vectors)进行转导,于转导72小时后获取其表达谱。另有一批EpiSC-5小鼠上胚层干细胞,采用携带针对多能性调控因子候选基因的短发夹RNA或非靶向对照的慢病毒载体进行转导,于转导72小时后分离mRNA,并使用PLATE-Seq技术(PMID:28740083)完成转录组分析。
