研究目的:通过RNA测序(RNA sequencing)表征内毒素休克与免疫耐受小鼠腹腔巨噬细胞的基因表达谱。 实验方法:通过腹腔灌洗分离小鼠腹腔巨噬细胞,经荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACS)筛选标记为F4/80+ CD11b+的细胞,提取内毒素休克与免疫耐受组小鼠腹腔巨噬细胞的RNA并进行RNA测序。 实验结果:内毒素休克与免疫耐受小鼠腹腔巨噬细胞呈现差异化基因表达特征。 研究结论:内毒素休克与免疫耐受小鼠腹腔巨噬细胞存在差异化基因表达。 整体实验设计:对经腹腔灌洗及荧光激活细胞分选(F4/80+ CD11b+)分离得到的小鼠腹腔巨噬细胞的低起始量RNA进行测序,每个样本混合2~3只小鼠的细胞。
