In a search for biomarkers for patients with cutaneous squamous cell carcinoma with high risk of metasitasis, a microarray comparison of miRNA expression profile between a non-metasitatic mouse keratinocyte cell line Pam212 and its metasitatic derivative LY-1 was carried out. Upregulation of the mir-221/222 cluster was the only notable change observed in LY-1 cells. Conluent cultures (single plate of each cell line) were compared. Signal-to-noise ratio was large enough and variation of duplicate spots were minimal for mir-221/222.
为筛选具有高转移风险的皮肤鳞状细胞癌患者的生物标志物,本研究通过微阵列技术对比了非转移性小鼠角质形成细胞系Pam212与其转移性衍生细胞系LY-1的microRNA(miRNA)表达谱。研究发现,LY-1细胞中仅miR-221/222簇出现显著上调。本次对比采用汇合状态的细胞培养物(每个细胞系仅设置单个培养板),且miR-221/222的信噪比足够高,重复点样的变异度极低。
