单细胞技术正处于快速迭代演进的阶段。本研究针对两项前沿单细胞表征技术——10x Genomics 3' GEMX与Illumina PIPseq V,开展性能基准测试,覆盖细胞类型鉴定与聚类推断两大核心性能维度。本研究结果可为单细胞技术的未来发展提供重要参考依据。
实验设计方案:从健康人类供体中分离外周血单个核细胞(peripheral blood mononuclear cells, PBMCs),将样本均分后,分别采用两种单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)技术——3' GEM-X与PIPseq v5进行平行实验处理。
