PAT-seq approach was utilised to determine the gene expression in the the transcriptome of C. auris treated with the vehicle DMSO or drug pyrvvinium pamoate (PP). Biological triplicates of C. auris cultures grown in RPMI medium with either DMSO or 1uM PP for 30 minutes at 37 Celsius were havested and total RNA was isolated using standard procedures (hot phenol method). 本研究采用PAT-seq方法(PAT-seq),对经溶剂二甲基亚砜(DMSO)或药物恩波吡维铵(pyrvvinium pamoate,PP)处理的耳念珠菌(Candida auris,C. auris)的转录组基因表达水平进行检测。收集三份生物学重复样本:耳念珠菌培养物分别于添加DMSO或1μM PP的RPMI培养基中,在37℃下培养30分钟后收集,并通过标准实验流程(热酚法)提取总RNA。实验整体设计:分析耳念珠菌中的poly(A)动态变化。
