MicroRNAs altered in colon adenomas

95 microRNA probes were found to have altered expression in colon adenomas compared to paired non-adenoma tissue Total mRNA was extracted from colon tissue that was flash frozen immediately after surgery. A total of 36 tissues were used for microarray analysis. Briefly, 5 ug of RNA were biotin labeled and hybridized to OSU-CCC microRNA microarrays version 4.0. We then analyzed associations comparing colon adenomas with paired non-adenoma tissue.

本研究发现，相较于配对的非腺瘤组织，结肠腺瘤样本中共有95个microRNA探针（microRNA probes）呈现表达差异。实验材料为术后即刻速冻的结肠组织，我们从中提取总mRNA，共计36份组织样本用于微阵列分析。简要流程如下：取5微克RNA进行生物素标记，随后与OSU-CCC第4.0版microRNA微阵列芯片进行杂交。后续我们开展关联分析，对比结肠腺瘤与配对的非腺瘤组织的表达差异。
