We here report ALKBH5, a m6A RNA demethylase, as a crucial oncogene in multiple myeloma (MM). Using various MM models, we demonstrated a critical requirement of ALKBH5 for MM cell proliferation in vitro and in vivo. 本研究报道ALKBH5——一种m6A RNA去甲基化酶(m6A RNA demethylase)——为多发性骨髓瘤(multiple myeloma,MM)中的关键致癌基因。借助多种多发性骨髓瘤模型,本研究证实ALKBH5对多发性骨髓瘤细胞的体外及体内增殖均具有关键调控作用。为鉴定ALKBH5的潜在mRNA靶标,本研究针对经ALKBH5敲低与未敲低的多发性骨髓瘤细胞中富集的mRNA样本开展了m6A测序(m6A-seq)。本研究还针对经短发夹RNA(shRNA)介导敲低ALKBH5与未敲低的RPMI-8226细胞开展了m6A测序。
