Host Factor Experiment (ECL001-R)

Purpose: To obtain samples from 2B-4 cells (a clonal derivative of Calu-3 cells) infected with MERS-CoV (previously called Hu CoV EMC virus) for transcriptional analysis. MERS-CoV and mock infected, the time points are 0, 3, 7, 12, 18 and 24h post-infection, except for mocks where the 18h was excluded. There are triplicate samples for all conditions where triplicates are defined as 3 different wells, plated at the same time using the same cell stock for all replicates; Time matched mocks done in triplicate from same cell stock as rest of samples. Inoculation medium for mock infection was the same as the medium used for virus infection. Infection done at an MOI of 5.

研究目的：获取感染中东呼吸综合征冠状病毒（MERS-CoV，曾被命名为Hu CoV EMC病毒）的2B-4细胞（Calu-3细胞的克隆衍生株）样本，用于转录分析。MERS-CoV感染组与模拟感染（mock infection）组的采样时间点均设为感染后0、3、7、12、18及24小时，但模拟感染组排除18小时时间点。所有实验条件均设置生物学重复样本，重复样本定义为同一批次接种、使用相同细胞储备液培养的3个独立孔板；与感染组时间匹配的模拟感染样本同样采用与其余实验组相同的细胞储备液制备，并完成3次生物学重复。模拟感染所用的接种培养基与病毒感染所用培养基完全一致；病毒感染的感染复数（Multiplicity of Infection, MOI）为5。
