In this study we employ NanoDam to identify targets within the epidermis for the transcription factor EFL-3 that we identified as a key epidermal development regulator. Overall design: Identification of EFL-3 genome-wide binding by epidermis-specific expression of GFP-nanobody:Dam fusion combined with EFL-3:GFP knock-in in C. elegans. Two different configurations of the GFP-nanobody:dam fusions are included (C-terminal and N-terminal fusions). Samples containing the GFP-nanobody:dam fusion only constitute control samples.

本研究采用NanoDam技术,针对我们已鉴定为表皮发育关键调控因子的转录因子EFL-3,鉴定其在表皮内的全基因组靶向结合位点。 实验整体设计:通过在秀丽隐杆线虫(C. elegans)中表皮特异性表达GFP纳米体(GFP-nanobody):Dam融合蛋白,并结合EFL-3:GFP基因敲入,实现对EFL-3全基因组结合位点的鉴定。 本研究包含两种GFP纳米体:Dam融合蛋白的构建形式:C端融合与N端融合。 仅表达GFP纳米体:Dam融合蛋白的样本作为对照样本。
