According to our analysis, PpRALF2 and 3 KO genotypes were more resistant to phytopathogens than wild type plants and PpRALF1 knockout lines. To expand our understanding of the roles of PpRALFs in biotic stress response, we next sought to perform RNA sequencing (RNA-seq) analysis to compare transcriptome response of PpRALF3 KO and wild type plants during F. solani infection. Twelve paired-end RNA-Seq libraries were generated from three biological replicates of mock-inoculated and F. solani-infected wild type and PpRALF3 KO plants.

本研究分析显示,PpRALF2与PpRALF3敲除(KO)基因型相较于野生型植株与PpRALF1敲除株系,对植物病原菌(phytopathogens)具有更强的抗性。为进一步阐明PpRALFs在生物胁迫响应(biotic stress response)中的功能,本研究后续开展了RNA测序(RNA-seq)分析,以对比茄病镰刀菌(F. solani)侵染过程中,PpRALF3 KO植株与野生型植株的转录组(transcriptome)应答差异。本研究从模拟接种(mock-inoculated)与F. solani侵染的野生型及PpRALF3 KO植株的3组生物学重复(biological replicates)中,构建了12个双端(paired-end)RNA-seq文库。
