We generated LNP-mRNAs specifically encoding wildtype (WT), B.1.351 and B.1.617 SARS-CoV-2 spikes, and systematically studied their immune responses. Single cell BCR-seq and TCR-seq unveiled repertoire diversity and clonal expansions in vaccinated animals. Overall design: Splenocytes were isolated and processed from immunized mice and analyzed by scVDJ-seq.

本研究构建了专门编码野生型(WT)、B.1.351株与B.1.617株严重急性呼吸综合征冠状病毒2(SARS-CoV-2)刺突蛋白的脂质纳米颗粒信使核糖核酸(LNP-mRNA),并系统研究了其诱导的免疫应答。单细胞B细胞受体测序(Single cell BCR-seq)与T细胞受体测序(TCR-seq)揭示了免疫动物体内的免疫组库多样性与克隆扩增特征。实验整体设计:从免疫小鼠体内分离脾脏淋巴细胞并完成处理,通过单细胞VDJ测序(scVDJ-seq)开展分析。
