RCOR2 and HDAC1 directly co-binds to CIITA gene. RCOR2 knockout does not affect HDAC1 binding on CIITA gene. RCOR2 has no binding on most MHCII genes. MDA-MB-231 cells overexpressing HA tagged RCOR2 were subjected to ChIP-seq with anti-HA antibody. Parental and RCOR2-KO MDA-MB-231 cells were subjected to ChIP-seq with anti-HDAC1 antibody. Occupancy of RCOR2 and HDAC1 on CIITA and MHCII genes were analyzed.

REST共抑制因子2(RCOR2)与组蛋白去乙酰化酶1(HDAC1)可直接共同结合于Ⅱ类主要组织相容性复合体反式激活因子(CIITA)基因。RCOR2基因敲除不会影响HDAC1在CIITA基因上的结合。RCOR2几乎不结合绝大多数主要组织相容性复合体Ⅱ类(MHCII)基因。对过表达血凝素标签(HA tag)RCOR2的MDA-MB-231细胞,采用抗HA抗体进行染色质免疫共沉淀测序(ChIP-seq);对亲本野生型及RCOR2基因敲除的MDA-MB-231细胞,采用抗HDAC1抗体进行ChIP-seq分析,并对RCOR2与HDAC1在CIITA基因及MHCII基因上的结合占有率进行了分析。
