We examined the gene expression profiles of liver samples throughout the day/night cycle (ZT0-ZT4-ZT8-ZT12-ZT16-ZT20) from control mice (night-restricted fed) and mice under short-term caloric restriction. Overall design: Bulk RNA sequencing (RNA-seq) from livers of WT and hepatocyte-deleted for Glucocorticoid Receptor mice (GR liver KO ) upon night restricted feeding (NR) and caloric restriction (CR) with sampling every 4hours at 6 different timepoints (ZT0, ZT4, ZT8, ZT12, ZT16, and ZT20), ZT corresponds to Zeitgeber Time, 6 a.m :ZT0 & ZT12: 6 p.m. Additionally, bulk RNA sequencing (RNA-seq) from livers of night-restricted fed (NR) and caloric restricted (CR) WT mice at Zeitgeber 12 (ZT12; 6 p.m).
本研究分析了昼夜周期(ZT0-ZT4-ZT8-ZT12-ZT16-ZT20)内,夜间限食对照组小鼠与短期热量限制小鼠的肝脏基因表达谱。总体实验设计:针对野生型(wild type,WT)和肝细胞糖皮质激素受体(Glucocorticoid Receptor,GR)敲除(GR liver KO)小鼠,分别在夜间限食(night-restricted fed,NR)与热量限制(caloric restriction,CR)条件下,于6个时间点(每4小时采样一次,即ZT0、ZT4、ZT8、ZT12、ZT16、ZT20)采集肝脏组织,开展批量RNA测序(bulk RNA sequencing,RNA-seq);其中ZT即授时因子时间(Zeitgeber Time),ZT0对应上午6时,ZT12对应下午6时。此外,本研究还对夜间限食(NR)与热量限制(CR)的野生型(WT)小鼠,在授时因子12(ZT12,即下午6时)时采集肝脏组织进行批量RNA测序。
