用于构建图2的数据集:为使用Seurat生成的聚类簇分配细胞系类型,该分析通过rCASC工具实现。
A) RNA-5c聚类:选取5000个高表达基因中变异度最高的2500个基因(通过rCASC的topx函数完成筛选),使用Seurat生成5个聚类簇(分辨率参数为0.1)。
B) RNA-3c聚类:沿用RNA-5c所用的2500个筛选基因,通过Seurat生成4个聚类簇(分辨率参数为0.1)。
C) RNA-5c层级聚类(采用欧氏距离、平均连锁法):对经行均值中心化处理的log2 CPM聚类伪批量表达数据(调用rCASC的bulkClusters函数),以及经行均值中心化处理的CCLE (Cancer Cell Line Encyclopedia)肺癌细胞系A449、NCIH838、NCIH2228、NCIH1975与HCC827的log2 TPM数据进行聚类分析。
