Neutrophils are known to be stimulated by different periodontal bacteria to produce reactive oxygen species and cytokines. It is inportant to investigate the gene changes made by bacteria of importance, of which, for periodontal disease, fusobaterium nucleatum is one. we used microarrays to investigate gene experssion changes in peripheral blood neutrophils werwhich e stimulated with or with out Fusobacterium Nucleatum (10953). Neutrophils from periodonatlly healthy individuals (n=4) were isolated and stimulated for 3hrs with or without fusobaterium nucleatum (10953). RNA was then extracted from these and pooled before hybridization on Affymetrix microarrays
中性粒细胞(Neutrophils)可被多种牙周细菌刺激产生活性氧(reactive oxygen species)与细胞因子(cytokines)。探究此类关键细菌诱导的基因表达变化具有重要意义,其中与牙周疾病相关的具核梭杆菌(Fusobacterium nucleatum)便是此类研究的目标菌种之一。本研究采用基因芯片(microarrays)技术,分析经具核梭杆菌(菌株编号10953)刺激与未刺激的外周血中性粒细胞(peripheral blood neutrophils)的基因表达变化情况。我们从牙周健康个体(n=4)中分离中性粒细胞,分别以具核梭杆菌(10953)刺激或不刺激,培养3小时。随后提取各组细胞的总RNA并混合,之后在Affymetrix基因芯片上完成杂交实验。
