To characterize porcine placental miRNAs, we performed small RNA-seq experiments using porcine placental tissues and the porcine placental trophoblast cell line pTr. To investigate the intracellular responses of pTr cells following JEV infection, RNA-seq and small RNA-seq experiments were conducted on pTr cells with or without JEV infection. Overall design: We collected placental samples from first - parity naturally delivered Duroc gilts, pTr cell samples under normal culture conditions, and pTr cell samples at 24 and 48 hours post - infection with JEV as well as non - infected samples. Two replicates per sample were used for RNA-seq and small RNA-seq experiments.
为解析猪胎盘微小RNA(miRNAs)的表达特征,本研究利用猪胎盘组织及猪胎盘滋养层细胞系pTr开展了小RNA测序(small RNA-seq)实验。为探究乙型脑炎病毒(JEV)感染pTr细胞后引发的细胞内应答反应,本研究对感染与未感染JEV的pTr细胞分别进行了RNA测序(RNA-seq)与小RNA测序实验。
整体实验设计:采集初产自然分娩杜洛克后备母猪的胎盘样本、正常培养条件下的pTr细胞样本,以及JEV感染后24小时、48小时的pTr细胞样本与未感染对照样本;所有样本均设置2个生物学重复,用于后续RNA-seq及small RNA-seq实验。
