Validation of an in vitro model of hypoxia using a primary cell culture of mouse embryonic fibroblasts (MEF) exposed to 1% oxygen at 37 degrees celsius. Control cells were exposed to atmospheric oxygen. Overall design: Hypoxic and normoxic MEF (in 100 mm dishes, four replicates per group) were submitted to gene expression analysis using Affymetrix Exon microarrays. Differential gene expression analysis was performed with R package DEMI (http://biit.cs.ut.ee/demi/).

本研究采用暴露于37℃、1%氧气环境的小鼠胚胎成纤维细胞(mouse embryonic fibroblasts, MEF)原代细胞培养物,对缺氧体外模型进行验证。对照组细胞则置于常压氧气环境中培养。总体实验设计:将缺氧组与常氧组MEF接种于100mm培养皿中,每组设置4个生物学重复,随后采用Affymetrix外显子微阵列(Affymetrix Exon microarrays)开展基因表达分析。差异基因表达分析使用R语言包DEMI完成,分析工具官方网址为http://biit.cs.ut.ee/demi/。
