mTORC2-deficient human embryonic stem cell (hESC) lines have been generated by disrupting RICTOR gene that encodes the specific subunit of mTORC2. Although RICTOR-knockout (RIC-KO) hESCs can maintain self-renewal and the expression of pluripotent markers, RIC-KO considerably impairs cell adhesion and cell-cell interaction in hESCs by disturbing RhoA/Rac signaling pathways. The difference of transcriptomic profiles between WT and RIC-KO hESCs needs to be further explored.
通过敲除编码雷帕霉素靶蛋白复合物2(mTOR complex 2, mTORC2)特异性亚基的RICTOR基因,已成功构建mTORC2缺陷型人类胚胎干细胞(human embryonic stem cell, hESC)株系。尽管RICTOR基因敲除(RIC-KO)的hESCs能够维持自我更新能力与多能性标志物的表达,但RIC-KO可通过干扰RhoA/Rac信号通路,显著损害hESCs的细胞黏附与细胞间相互作用。野生型(WT)与RIC-KO hESCs之间的转录组图谱差异仍有待进一步探究。
