This study employs environmental DNA (eDNA) metabarcoding to characterize the plant community within a tropical campus environment on Hainan Island, China. Environmental samples (e.g., soil) were collected from various locations across the campus to capture the local floristic diversity. Total DNA was extracted from these samples, and two complementary plant-specific barcode regions, rbcL and ITSu, were amplified via polymerase chain reaction (PCR) to maximize taxonomic resolution and coverage. The resulting amplicon libraries were sequenced on an Illumina MiSeq platform to generate high-throughput sequencing data.

本研究采用环境DNA(eDNA)元条形码测序技术,对中国海南岛某热带校园环境中的植物群落进行表征。研究人员从校园内多个点位采集了土壤等环境样本,以覆盖当地植物区系多样性。从上述样本中提取总DNA后,通过聚合酶链式反应(PCR)扩增rbcL与ITSu这两个互补的植物特异性条形码区域,以最大化分类学分辨率与覆盖范围。最终得到的扩增子文库在Illumina MiSeq测序平台上完成测序,生成高通量测序数据。
