RNA-seq was performed to look at global differences in gene expression between human embryonic stem cells (hESCs) particularly H1 cell line (WA-01, passage 23-40) cultured in distinct conditions. In details we compared cells adapted to primed (mTeSR1) and two naïve states: feeder dependent (4iLA + feeder) and feeder free (FINE) states.
本研究通过RNA测序(RNA-seq)探究人类胚胎干细胞(human embryonic stem cells, hESCs)在不同培养条件下的全基因组基因表达差异,实验所用细胞为特定的H1细胞系(WA-01,传代23-40代)。具体而言,本研究比较了适应始发态(primed)培养体系(mTeSR1培养基)的细胞,以及两种初始态(naïve)干细胞:饲养层依赖型(4iLA + 饲养层细胞)和无饲养层型(FINE培养体系)的细胞。
