This study aims to examine changes to the microRNA biogenesis pathway when the exonuclease Dicer is mutated such that it cannot recruit its RNA-binding partner proteins, TRBP and PACT. These partner proteins may contribute to changes in the processing of microRNAs, including effects on microRNA length and their loading onto targeting effector protein Ago2. The study used immunoprecipitation of Ago2 followed by sequencing of associated microRNAs to text for differences between cells containing Dicer that could recruit TRBP/PACT and those that could not

本研究旨在探究当核酸外切酶Dicer发生突变,无法招募其RNA结合伴侣蛋白TRBP与PACT时,微小RNA(microRNA)生物发生通路的变化。此类伴侣蛋白或可对微小RNA的加工过程产生影响,包括调控微小RNA的长度及其加载至靶向效应蛋白Ago2的过程。本研究采用Ago2免疫沉淀联合关联微小RNA测序的方法,对可招募TRBP/PACT的Dicer细胞与无法招募该伴侣蛋白的细胞之间的差异进行检测。
