Effect of FSCN1 gene inactivation in the H295R cell line

The actin-bundling protein fascin (FSCN1) is overexpressed in aggressive adrenocortical carcinoma (ACC) and represents a reliable prognostic indicator. We investigated the effects of FSCN1 inactivation by CRISPR/Cas9 in ACC H295R cells on global gene expression profiles in those cells. We performed gene expression profiling analysis using data obtained from RNA-seq of 2 different H295R control clones and 2 different FSCN1 KO clones (2 biological replicates for each clone). Comparative gene expression profiling analysis of RNA-seq data for control and FSCN1 KO H295R clones.

肌动蛋白束蛋白fascin（FSCN1）在侵袭性肾上腺皮质癌（ACC）中呈高表达，且是可靠的预后标志物。本研究探究了CRISPR/Cas9介导的FSCN1敲除对ACC来源H295R细胞全局基因表达谱的影响。我们针对2株独立的H295R对照克隆及2株独立的FSCN1敲除（KO）克隆的RNA测序（RNA-seq）数据开展基因表达谱分析，每个克隆设置2个生物学重复。本研究对对照与FSCN1敲除H295R克隆的RNA测序数据进行了比较基因表达谱分析。
