To investigate the characteristics of PD-1+ LSCs in T-ALL, we purified PD-1+ and PD-1– T-ALL cells from the bone marrow (femur and tibia) of T-ALL mice, Pten null T-ALL mice, or PDX recipients. RNA sequencing was performed to explore the differential gene expression and pathway enrichment of PD-1+ LSCs in T-ALL. Overall design: Comparative gene expression profiling analysis and gene set enrichment analysis of RNA-seq data for PD-1+ LSCs in murine and human T-ALL.
为探究T细胞急性淋巴细胞白血病(T-cell Acute Lymphoblastic Leukemia, T-ALL)中PD-1阳性白血病干细胞(PD-1+ Leukemia Stem Cells, PD-1+ LSCs)的特性,我们从T-ALL小鼠、Pten缺陷型T-ALL小鼠或患者来源异种移植(Patient-derived Xenograft, PDX)受体的骨髓(股骨与胫骨)中纯化得到PD-1阳性与PD-1阴性的T-ALL细胞。随后开展RNA测序(RNA-seq),以解析T-ALL中PD-1+ LSCs的差异基因表达与通路富集情况。
整体实验设计:针对小鼠与人类T-ALL来源的PD-1+ LSCs的RNA测序数据,开展对比基因表达谱分析与基因集富集分析(Gene Set Enrichment Analysis, GSEA)。
