我们提出,HS2可通过激活邻近的CCCTC结合因子(CCCTC-binding factor,CTCF)结合位点,参与β-珠蛋白拓扑相关结构域(Topologically Associating Domain,TAD)的构建,且该功能超越了典型增强子的活性范畴。实验设计:在对照组及基因座控制区(Locus Control Region,LCR)HS2缺失细胞中,分别开展组蛋白H3赖氨酸27乙酰化(histone H3 lysine 27 acetylation,H3K27ac)、组蛋白H3赖氨酸27三甲基化(histone H3 lysine 27 trimethylation,H3K27me3)的染色质免疫共沉淀测序(Chromatin Immunoprecipitation sequencing,ChIP-seq)实验。
