In order to identify the targets of miR-193a-5p in osteosarcoma U2OS cell line, we used a lentivirus-mediated expression system to overexpressing miR-193a precusor, miR-193a-5p target sequence and non-target sequence, respectively, in osteosarcoma cell line U2OS. A tandem mass tag (TMT)-based quantitative proteomic strategy was employed to identify the global profile of miR-193a-5p-regulated proteins. order to identify the targets of miR-193a-5p, we used a lentivirus-mediated expression system to overexpressing miR-193a precusor, miR-193a-5p target sequence and non-target sequence, respectively, in osteosarcoma cell line U2OS. 为鉴定骨肉瘤U2OS细胞系中微小RNA-193a-5p(miR-193a-5p)的靶标,我们采用慢病毒介导的表达系统,分别在骨肉瘤U2OS细胞系中过表达miR-193a前体、miR-193a-5p靶序列及非靶序列。随后运用基于串联质量标签(Tandem Mass Tag, TMT)的定量蛋白质组学策略,以鉴定miR-193a-5p调控的蛋白质全局表达谱。
