Major transcriptional changes (300 differentially expressed genes) take place upon downregulation of Lamin A/C in B16F10 melanoma cells Overall design: Lamin A and lamin C expression in the nuclei of the B16F10 melanoma cells were downregulated by stable introduction of a lamin A/C shRNA construct., resulting in an expected increase in nuclear deformability and squeezing capacities in our melanoma cell model. RNA was isolated from both shControl and shLmna B16F10 cells for transcriptome analysis via MARS-seq sequencing and bioinformatics analysis.

在B16F10黑色素瘤细胞中下调核纤层蛋白A/C(Lamin A/C)的表达后,会发生显著的转录组改变,涉及300个差异表达基因。整体实验设计:通过稳定转染靶向核纤层蛋白A/C的短发夹RNA(shRNA)构建载体,下调B16F10黑色素瘤细胞核内的核纤层蛋白A与核纤层蛋白C的表达水平,该操作可预期提升本黑色素瘤细胞模型的细胞核变形能力与挤压能力。随后分别从对照短发夹RNA(shControl)转染与靶向核纤层蛋白A/C的短发夹RNA(shLmna)转染的B16F10细胞中提取RNA,通过MARS-seq测序及生物信息学分析开展转录组学研究。
