Purpose:The differences of host regulatory networks mediated by different generations of PEDV strains were explored by transcriptome sequencing.Method:The high generation PEDV-infected group(P120), low generation PEDV-infected group(P16) and mock-infected group received samples at 12, 24 and 36 hours post infection, and each group had three replicates at each time point.Conclusion: our study is the first to analyze the changes of gene transcription level of host cells caused by different generations of PEDV infection by using the pig intestinal organoid model. It provides a new strategy for the prevention and treatment of PEDV.

研究目的:本研究采用转录组测序技术,探究不同传代代次的猪流行性腹泻病毒(Porcine Epidemic Diarrhea Virus, PEDV)毒株所介导的宿主调控网络差异。 研究方法:设立高代次PEDV感染组(P120)、低代次PEDV感染组(P16)及空白对照组,分别于感染后12、24、36小时采集样本,每组在各时间点均设置3个生物学重复。 研究结论:本研究首次利用猪肠道类器官模型,解析不同代次PEDV感染宿主细胞后诱导的基因转录水平变化,为猪流行性腹泻病毒的防治提供了全新策略。
