Double mutant heat labile (dmLT) adjuvant is being proposed for oral vaccine formulation. dmLT stability in saliva was evaluated by dmLT ELISA. dmLT stability was tested using two separate preparation methods to prevent any interference with saliva and the ELISA method. In the first method dmLT was mixed with saliva in tubes, incubated at 37˚C, and tested at 5, 10, 30 minutes by ELISA. In the second method saliva was added to dmLT previously bound to ELISA plates. Plates were tested at the same specified time points.
双突变热不稳定佐剂(double mutant heat labile, dmLT)被提出用于口服疫苗制剂的研发。本研究通过dmLT特异性酶联免疫吸附试验(ELISA)评估了该佐剂在唾液中的稳定性。为避免唾液与ELISA检测方法之间产生相互干扰,研究采用两种独立的样本制备方法对dmLT的稳定性进行了检测。第一种方法:将dmLT与唾液在试管中混合,于37℃条件下孵育,分别在5、10、30分钟时通过ELISA进行检测。第二种方法:将唾液添加至预先结合于ELISA酶标板的dmLT体系中,于相同的指定时间点对酶标板进行检测。
