To investigate the function of H3K9me3 in constitutive heterochromatin formation in early embryo we have created embryos that are devoid of H3K9me3 Overall design: different stages of embryos of control, single mutants and triple mutant were subjeted to Cut and tag for H3K9me2/3, HP1a, and H3.
为探究组蛋白H3赖氨酸9位三甲基化修饰(H3K9me3)在早期胚胎组成型异染色质形成过程中的功能,我们构建了完全缺失H3K9me3的胚胎。实验设计:对对照组、单突变体及三突变体的不同发育阶段胚胎,开展针对组蛋白H3赖氨酸9位二甲基化/三甲基化修饰(H3K9me2/3)、异染色质蛋白1a(HP1a)以及组蛋白H3的CUT&Tag(Cut and Tag)实验。
