We analyze gene regulatory mechanisms involved in adrenocortical steroidogenesis using assays to measure RNA expression, synthesis, decay, processing, translation and miRNA expression. Overall design: Metabolic labeling, ribosome profiling and small RNA-sequencing was performed on human H295R adrenocortical cell lines with and without treatment with angiotensin or forskolin. RNA-seq was performed on adrenal glands of p15 c57bl/6 mice.

本研究通过开展检测RNA表达、合成、降解、加工、翻译及microRNA (miRNA)表达的相关实验,分析参与肾上腺皮质类固醇生成的基因调控机制。整体实验设计:在经血管紧张素或佛司可林处理与未处理的人源H295R肾上腺皮质细胞系中,开展代谢标记、核糖体谱分析及小RNA测序实验;同时对p15日龄C57BL/6小鼠的肾上腺组织开展RNA测序(RNA-seq)实验。
