Centaurea cyanus (XX-0-BRAUN-7665519) seeds were obtained from the Botanical Garden at TU Braunschweig. Plants were grown under long day conditions (16h light/8h darkness) at room temperature. High molecular weight genomic DNA was extracted with a CTAB-based protocol (https://dx.doi.org/10.17504/protocols.io.bcvyiw7w). Short DNA fragments were depleted using the SRE kit. Library preparation for nanopore sequencing was performed with SQK-LSK114. Sequencing was performed on R10 flow cells in MinION Mk1B. Super high accuracy basecalling was performed with Guppy (v6.5.7+ca6d6af).

本实验所用矢车菊(Centaurea cyanus,编号XX-0-BRAUN-7665519)的种子取自布伦瑞克工业大学(TU Braunschweig)植物园。植株于长日照培养条件(16小时光照/8小时黑暗)下在室温环境中培养。采用基于十六烷基三甲基溴化铵(CTAB)的提取方案(详见https://dx.doi.org/10.17504/protocols.io.bcvyiw7w)提取高分子量基因组DNA。使用SRE试剂盒去除样本中的短DNA片段。采用SQK-LSK114试剂盒完成纳米孔测序文库构建。测序实验在搭载R10流动池的MinION Mk1B测序平台上开展。使用Guppy(v6.5.7+ca6d6af)完成超高精度碱基识别(basecalling)。
