Profiling circular RNAs in HeLa cell

We used RNA-seq to examine circular RNAs from RNase R treated ribo-zero RNAs in HeLa cells. HeLa total RNA was supplied in the Ion Total RNA-Seq Kit v2, 12-Reaction Kit (Life technologies, Cat. no. 4475936) for preparing cDNA library. cDNA library preparation and sequencing were performed on Ion Torrent, and preliminary data acquisition and preprocessing was performed in house.

我们采用RNA测序（RNA-seq）技术，对海拉细胞（HeLa cells）中经核糖核酸酶R（RNase R）处理的核糖体去除RNA（ribo-zero RNAs）内的环状RNA（circular RNAs）开展检测分析。本实验用于构建互补DNA（cDNA）文库的海拉细胞总RNA，取自Ion总RNA测序试剂盒v2 12反应装（Life technologies，货号：4475936）。互补DNA文库的制备与测序均在离子激流（Ion Torrent）测序平台上完成，初步数据的获取与预处理工作由本机构内部完成。
