To investigate the RNA differences in muscle with and without constitutive AHR activation. AAV9 was used to express either a green fluorescent protein (control) or mutant AHR protein that exhibits constitutive transcriptional activity Overall design: Male C57BL6J mice were infected with AAV9 viruses encoding a GFP or constitutively active AHR driven by the human skeletal actin promoter to cause myofiber-specific expression. AAV's were injected into the gastrocnemius mucle of 5-month old mice and muscles were harvested 5 months later for RNA isolation

本研究旨在比较组成型芳香烃受体(aryl hydrocarbon receptor, AHR)激活与未激活状态下小鼠肌肉组织的RNA表达差异。实验采用腺相关病毒血清型9(adeno-associated virus serotype 9, AAV9)介导表达绿色荧光蛋白(green fluorescent protein, GFP,作为对照),或具有组成型转录活性的突变型AHR蛋白。整体实验设计如下:将由人骨骼肌肌动蛋白启动子驱动、可实现肌纤维特异性表达的编码GFP或组成型激活型AHR的AAV9病毒,接种于5月龄雄性C57BL/6J小鼠的腓肠肌中;于病毒接种5个月后采集肌肉组织,用于RNA分离提取。
