The research is dedicated to the study of anticancer activity of two live S. pyogenes strains. The study aimed to investigate the oncolytic efficacy of S. pyogenes GUR and its M protein knockout mutant, S. pyogenes strain GURSA1, which has been genetically constructed to minimize overall toxicity, against mouse hepatoma 22A, pancreatic cancer PANC02, and human glioma U251 cells, both in vitro and in vivo, using the C57BL/6 mouse model. Using the C57BL/6 mouse model of pancreatic cancer, we also demonstrated that the oncolytic S. pyogenes GURSA1 strain selectively colonized tumors 24 hours after intraperitoneal administration, while the strain was not detected in the spleens or livers of the animals.
本研究聚焦于两株活的化脓性链球菌(Streptococcus pyogenes,简称S. pyogenes)的抗癌活性探究。本研究旨在考察化脓性链球菌GUR株及其经基因工程构建以降低整体毒性的M蛋白敲除突变株——化脓性链球菌GURSA1株,针对小鼠肝癌22A、胰腺癌PANC02以及人类胶质瘤U251细胞的溶瘤效力,实验覆盖体外(in vitro)与体内(in vivo)两种研究体系,并采用C57BL/6小鼠模型。此外,本研究借助胰腺癌C57BL/6小鼠模型进一步证实:溶瘤性化脓性链球菌GURSA1株在腹腔注射24小时后可选择性定植于肿瘤组织,且未在受试动物的脾脏与肝脏中检出该菌株。
