We report the binding profile of A. thaliana CENH3, L. oleraceum CENH3 and Z. mays CENH3 in the genetic background of A. thaliana Col ecotype. The L. oleraceum CENH3 and Z. mays CENH3 ChIP was performed in a cenh3 homozygous mutant background expressing CENH3 homologs as transgenes. Overall design: Examination of 3 different centromeric histone H3 (CENH3) variants in 3 different A. thaliana genotypes.

本研究报道了拟南芥(Arabidopsis thaliana)Col生态型遗传背景中,拟南芥CENH3、欧独行菜(Lepidium oleraceum)CENH3与玉米(Zea mays)CENH3的结合谱。本研究中,欧独行菜CENH3与玉米CENH3的染色质免疫共沉淀(Chromatin Immunoprecipitation,ChIP)实验,在表达CENH3同源转基因的cenh3纯合突变体遗传背景内完成。实验整体设计:在3种不同的拟南芥基因型中,检测3种不同的着丝粒组蛋白H3(centromeric histone H3,CENH3)变体。
