The Rutidosis leptorrhynchoides (also spelled R. leptorhynchoides, Button Wrinklewort) genome project aims to produce a high-quality, telomere-to-telomere assembly from one diploid individual plants of Truganina Cemetery ancestry to support conservation efforts for this endangered south-eastern Australian species, threatened by habitat loss and fragmentation and genetic erosion. Plant 2 (leaves) was used for HiC, and long read sequencing on the PacBio Revio and ONT PromethION platforms. A second plant, Plant 1, was used for Illumina RNASeq (leaf, & stem). This project provides essential resources to improve our understanding of the species' genomic architecture, including self-incompatibility mechanisms crucial for reproduction. These resources will contribute to a broader study developing genomic data to inform genetic management strategies, including assisted gene flow between isolated populations. By enhancing genetic diversity and adaptive potential, this research aims to ensure the long-term survival and resilience of R. leptorrhynchoides in the face of environmental change.
