Polycomb group (PcG) proteins maintain cell identity by transcriptionally silencing developmental genes during embryogenesis. Phc1 is a subunit of cPRC1 known for gene repression, in the maintenance of pluripotency. To further explore how Phc1 might regulate the pluripotency of mESCs in both PRC1-dependent and PRC1-independent manners, we performed RNA-sequencing of the wildtype, Phc1-/- and Phc1-/- + Nanog overexpression mESCs.
多梳蛋白家族(Polycomb group, PcG)可在胚胎发生过程中通过转录沉默发育基因以维持细胞身份。Phc1是经典多梳抑制复合体1(canonical Polycomb repressive complex 1, cPRC1)的亚基,已知其在多能性维持中介导基因沉默功能。为进一步探究Phc1如何以PRC1依赖性与非依赖性两种方式调控小鼠胚胎干细胞(mouse embryonic stem cells, mESCs)的多能性,本研究对野生型、Phc1纯合敲除型以及Phc1纯合敲除并过表达Nanog的小鼠胚胎干细胞开展了RNA测序(RNA-sequencing)分析。
