Notochordal cells are considered as a rare cell type ensuring signaling function required for proper development and homeostasis in various tissues. We used single cell RNA sequencing (scRNA-seq) to characterize gene signature of notochordal-like cells and understand heterogeneity of co-emerging cell types during in vitro differentiation. Overall design: hiPS-NLC differentiating cells were dissociated and labelled with cell-multiplexing oligos (CMO) according to hiPSC line origin and treatment and analyzed using scRNAseq.

脊索细胞(Notochordal cells)被认为是一类稀有细胞类型,其介导的信号功能是多种组织正常发育与稳态维持所必需的。本研究采用单细胞RNA测序(scRNA-seq)对脊索样细胞的基因特征进行解析,并探究体外分化过程中共同出现的细胞类型的异质性。实验设计概述:我们根据人诱导多能干细胞系的来源及处理方式,对hiPS-NLC分化细胞进行解离,并用细胞多重标记寡核苷酸(CMO)进行标记,随后通过scRNA-seq开展测序分析。
