Nowadays, sepsis and septic shock have become major public health problems, which are the main cause of death among patients admitted to the intensive care units. Notably, myocardial dysfunction during sepsis, usually called sepsis-induced myocardial depression, is common in septic shock patients whose incidence is about 70% and leads to a high mortality. However, the mechanism underlying the septic myocardial depression is still unclear. Recent studies suggested noncoding RNAs (ncRNAs) could regulate the process of sepsis-induced myocardial depression. But the role of transcriptome in myocardial depression is unknown, and all kinds of ncRNAs as well as messenger RNAs (mRNAs) present in sepsis-induced myocardial depression have never been sequenced. Therefore, we performed the rat models of sepsis-induced myocardial depression and investigated ncRNAs and mRNAs expression profiles in heart tissue between sepsis and control group using whole-genomic next-generation sequencing.

当前,脓毒症(sepsis)与脓毒性休克(septic shock)已成为重大公共卫生问题,亦是重症监护病房(intensive care units)收治患者的主要死亡原因。值得注意的是,脓毒症病程中出现的心肌功能障碍(myocardial dysfunction)通常被称为脓毒症诱导的心肌抑制(sepsis-induced myocardial depression),在脓毒性休克患者中发生率极高,约为70%,且可导致较高的病死率。然而,脓毒症诱导的心肌抑制的潜在分子机制至今仍未阐明。既往研究提示,非编码RNA(noncoding RNAs, ncRNAs)可调控脓毒症诱导的心肌抑制的发生发展过程,但转录组(transcriptome)在该心肌抑制过程中的作用尚不明确,且脓毒症诱导的心肌抑制状态下存在的各类非编码RNA与信使RNA(messenger RNAs, mRNAs)尚未被系统性测序分析。为此,本研究构建了脓毒症诱导的心肌抑制大鼠模型,并采用全基因组下一代测序(whole-genomic next-generation sequencing)技术,对比分析了脓毒症组与对照组大鼠心脏组织中的ncRNAs与mRNAs表达谱。
