Single-cell RNA sequencing (scRNA-seq) was performed on the CD31-mircrobeads enriched endothelial cells isolated from aorta and heart from eight-week-old, male Ldlr-/- mice fed standard chow (Chow) or diabetogenic high-fat diet with 0.15% cholesterol (DDC) diet for 12 weeks. This study report the changes of EC subpopulations, fractions, transcriptomic and metabolic profiles during diabetic atherosclerosis. Overall design: Examination of the transcriptomic profiles of endothelial cells isolated from aorta and heart pooled from three Ldlr-/- mice fedstandard chow (Chow) or diabetogenic high-fat diet with 0.15% cholesterol (DDC) diet for 12 weeks

本研究对8周龄雄性Ldlr⁻/⁻小鼠的主动脉与心脏来源、经CD31-mircrobeads富集的内皮细胞开展单细胞RNA测序(Single-cell RNA sequencing,scRNA-seq),实验小鼠分为两组,分别饲喂标准饲料(Chow)或含0.15%胆固醇的致糖尿病高脂饮食(DDC diet),干预时长为12周。本研究报道了糖尿病性动脉粥样硬化进程中内皮细胞亚群、细胞占比、转录组及代谢谱的变化。整体实验设计:对三只分别饲喂标准饲料(Chow)或含0.15%胆固醇的致糖尿病高脂饮食(DDC diet)12周的Ldlr⁻/⁻小鼠,取其主动脉与心脏组织中分离得到并混合的内皮细胞,对其转录组谱进行检测。
