The greater amberjack (Seriola dumerili) is an economically significant cosmopolitan species, which distributedin the open sea andnearshore reef habitats. As a fast-growing species, it holds outstanding swimming ability and abundant meat-based nutrients, which make appropriatefor marine ranching construction and marketing purposes. However, the incomplete of high-quality genome has limited its conservation, molecular research, and evolution, especially genetic resources for selection breeding. Here, Pacific Biosciences (PacBio) High-Fidelity (HiFi) sequencing, Hi-C chromatin conformation capture,and Oxford Nanopore Technologies (ONT) ultra-long readswere utilized assemble the telomere-to-telomere gap-free genome of greater amberjack.

高体鰤(Seriola dumerili)是一种具有重要经济价值的广布性海洋物种,广泛分布于开阔海域及近岸礁盘生境中。作为生长快速的物种,其兼具优异的游泳能力与丰富的肉质营养成分,非常适合用于海洋牧场建设与商业化开发。然而,目前高质量参考基因组的不完备性,限制了该物种的保护生物学、分子研究及演化生物学研究,尤其是选择育种所需遗传资源的挖掘与利用。本研究利用太平洋生物科学(Pacific Biosciences, PacBio)高保真(High-Fidelity, HiFi)测序技术、Hi-C染色质构象捕获技术,以及牛津纳米孔科技(Oxford Nanopore Technologies, ONT)超长读长测序技术,成功组装出高体鰤的端粒到端粒无间隙参考基因组。
