DHAV-1 infects DEF and miRNAs and mRNA of ducklings.We characterized and compared miRNA and mRNA expression profiles in duck embryonic fibroblasts (DEF) and the liver of ducklings infected with DHAV-1. 130 and 72 differentially expressed (DE) miRNA (DEM),2204 and 1976 differentially expressed (DE) mRNA (DEG) were identified. 15 up-regulated DEM, 5 down-regulated DEM,340 up-regulated DEG, and 50 down-regulated DEG were found in vitro and in vivo DHAV-1 infection. In particular, we identified target genes and functional annotations of DE mRNA in vivo and in vitro infections. We enriched with multiple gene ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, which may have important roles in viral virulence, cell proliferation, and metabolism

鸭甲型肝炎病毒1型(DHAV-1)可感染鸭胚成纤维细胞(Duck Embryo Fibroblasts, DEF)与雏鸭,并影响雏鸭的microRNA(miRNA)及信使RNA(mRNA)表达。本研究对感染DHAV-1的鸭胚成纤维细胞与雏鸭肝脏中的miRNA和mRNA表达谱进行了系统表征与比较。研究共鉴定得到130个与72个差异表达miRNA(DEM),以及2204个与1976个差异表达mRNA(DEG)。在DHAV-1的体外与体内感染模型中,发现15个上调差异表达miRNA、5个下调差异表达miRNA、340个上调差异表达mRNA与50个下调差异表达mRNA。尤为关键的是,本研究鉴定了体内外感染模型中差异表达mRNA的靶基因并完成了功能注释。我们对多条基因本体(Gene Ontology, GO)及京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes, KEGG)通路进行了富集分析,上述通路可能在病毒毒力、细胞增殖与代谢过程中发挥重要作用。
