Sequencing and characterization of the Atlantic bluefin tuna transcriptome aimed at SNP discovery and providing a novel genomic resource. Pooled cDNA libraries constructed with RNA extracted from 6 adult individuals and 14 developmental stages were sequenced on a an Illumina HiSeq2000. The 215 millions of paired-end reads obtained were assembled in a draft transcriptome comprised of 37,590 contigs ranging in size from 201 to 27,825 bp. SNP calling resulted in the identification of 147,534 SNPs. The genomic resource obtained will enable development of targeted assays for large numbers of polymorphic genetic markers.
大西洋蓝鳍金枪鱼转录组的测序与表征研究,旨在开展单核苷酸多态性(Single Nucleotide Polymorphism,SNP)发掘,并构建新型基因组资源。本研究使用从6尾成年个体及14个发育阶段样本中提取的RNA构建混合cDNA文库,依托Illumina HiSeq2000测序平台完成测序。所获2.15亿条双端读段经组装后,得到一套草稿转录组,包含37590个重叠群(contig),序列长度区间为201 bp至27825 bp。经SNP位点检测分析,共鉴定得到147534个SNP位点。本研究获得的基因组资源,将为开发针对大量多态性遗传标记的靶向检测方法提供支撑。
